Warning : This may not be suitable for all. Follow these steps if you want your system to be lightning fast.

Disclaimer : The list is not complete. I came up with this list from the experience i got from working with my 5 year old machine with 512 MB Ram and partially working motherboard.

Disable Visual Effects
Right click on My Computer, select Properties, go to Advanced -> select Settings under Performance -> Select Adjust for best performance.

Set virtual memory to double of your RAM. Keep the Initial and Maximum size same.

Keep the size of your C: drive (drive where you install Windows) at 10 GB

Install all programs in D:\Program Files\

Disable sounds (control panel)

Disable system restore

Disable Error Reporting

Disable Remote Assistance and Remote Desktop if you do not use them

Remove Desktop background

Remove all unwanted startup programs

Disable all unwanted services.

Uninstall all unwanted programs.

Defragment all disks

Disable Desktop Cleanup Wizard

Uncheck “Allow Indexing Service to index this disk for fast file searching” after right clicking each drive.

Run CheckDisc once in a while

Unshare all useless shared folders and files.

Uncheck hide inactive icons.

Run diskcleanup once in a while

Delete system restore files

Remove Windows Messenger

Use classic start menu

Disable hibernation if you are not using that. NB: Hibernation is a better method than sleep for saving time.
